Are you set on heading west? One summer I rode the Blue Ridge Parkway south into NC and crossed into Tenn. over the Cherohala Skyway. The only reason I avoided Tail of the Dragon was due to the weather conditions on the day I made it there. I'm like you, don't like to stay in one place during my trips, and I can tell you the experience of taking the BRP is worth it.
You can't do it all in one day, so depending on how much you want to ride I'd suggest stops in Asheville and Roanoke. I stayed in a nice motel in Chantilly, VA that's about 20 minutes from the Smithsonian in DC and the Discovery Space Shuttle. The back roads of PA are great and rt. 555 (triple nickel) just north of Marietta, OH is awesome!
The only west trip I took was through Indiana and Illinois so I don't have much advise that way. I remember the rain and many many detours to avoid storms, there's no way but through when you travel west. Rt. 67/52 north from Clinton, IA to Dubuque is a twisty dream at 60mph and follows the Mississippi, that was fun.
